Are you an adult who has asthma, or are you willing to be a healthy volunteer? This study is looking at the DNA in airway cells to see how they differ between people who have asthma and people who do not. To get a sample of cells from your airways, you may have a bronchoscopy (a tube inserted through your mouth into your airways). To participate in this study, you must be between 18 and 70 years old, either with asthma or with no history of asthma. This study is located in San Francisco, California.

Do you have asthma and obesity and live near Aurora, Colorado? Obesity can lead to widespread inflammation that may make asthma worse. This study will test whether a medicine called CXA-10 reduces inflammation in the airways and body and improves asthma symptoms in people who have obesity. To participate in this study, you must be between 18 and 65 years old, have a BMI of 30 or greater, and have been on long-acting controller medicine—such as inhaled corticosteroids or long-acting beta2-agonists—for at least three months. This study is located in Aurora, Colorado.

Are you an adult with both asthma and obesity? This study is testing whether a medicine called L-citrulline can help treat asthma and improve lung function in people who have obesity. To participate in this study, you must be 18 to 65 years old and have asthma and obesity. This study takes place in Durham, North Carolina.

Has your child been treated for an asthma attack with an oral corticosteroid medicine in the last 12 months? A study team will recruit 60 teens and young adults (between 12 and 21 years old) with persistent asthma who are at high risk for future exacerbation. Sickle Cell Disease (SCD) impairs oxygen transport to tissue and causes endothelial injury. Thus, therapeutic interventions aim to improve both, but there is an unmet need for biomarkers to determine when intervention is necessary and evaluate the effectiveness of the chosen intervention in individual patients. This study proposes to monitor SCD and its treatment through their impact on cerebral hemodynamics, as the brain is one of the most vulnerable and consequential targets of the disease. Specifically, this study will optimize quantitative magnetic resonance imaging (MRI) and advanced optical spectroscopy techniques such as frequency-domain near-infrared and diffuse correlation spectroscopies (FDNIRS-DCS) to monitor 1) cerebral oxygen transport with measures of cerebral blood flow (CBF), cerebral oxygen extraction fraction (OEF) and cerebral metabolic rate of oxygen consumption (CMRO2) and 2) endothelial function with cerebrovascular reactivity (CVR).

Are you or your child at least 13 years old and have sickle cell disease? In this stidy, researchers are trying to find out if a new medication improves the success of a blood stem cell transplant. People 13 years old or older who are eligible for a blood stem cell transplant to treat sickle cell disease may be able to participate. Healthy family members over age 13 who are matched to transplant recipients are also needed to donate blood for the study.
